Peter Guengerich Center in Molecular Toxicology, Department of Biochemistry, Vanderbilt University School of Medicine, Nashville, TN 37232-0146 USA Abstract Cytochrome P-450 ( P - 4 5 0 ) oxidations, although apparently diverse in their appearance, can be considered in terms of mechanisms involving abstraction of electrons or hydrogen atoms by hypervalent F e = 0 species and subsequent radical recombination.
Exciting results have been obtained using the expression system described ( 2 3 ) that permit the synthesis o f a single P - 4 5 0 or a combination of P-450s in cells devoid of this hemoprotein. This offers the means to establish "designer constructed" cells of predicted enzyme composition for the examination of integrated pathways o f metabolism. With the P - 4 5 0 enzymes, this approach offers the opporĀ tunity for developing new types of tests for toxicity evaluation o f drugs and other chemicals as well as the expression of large amounts of enzyme for commercial application to new techniques of chemistry.
